چکیده

A total of 1,110 decamer primers were screened for RAPD markers linked to a dominant allele in hazelnut (Corylus avellcma) that confers resistance to eastern filbert blight caused by A~zisogrcrinmn anomnl~z. Twenty R 4 P D markers linked in coupling, and five markers linked in repulsion. were found. A seedling population was used to construct a linkage map of the region flanlung the resistance locus. The map spans 46.6 ch1, with 1 1 markers on one side of the resistance locus and eight on the other side. Eleven markers showed less than 3% recombination with resistance, including three that showed no recombination. Seven of these 11 markers are sufficiently robust to allow their use in marker-assisted selection. These include XX138i,) which shows no recornbination, and six markers on one side of the resistance locus: 17350~), 152Roo, l?3Y25. 275,,;0, H19hsi, and 016,250. Marker 268580, which flanks the resistance locus on the other side, is also suitable for use in marker-assisted selection. but shows 5.8% recombination with resistance. Other markers are less suitable for marker-assisted selection because of sensitivity to changes in primer or kfsC12 concentration, or the long time required for electrophoresis to separate bands of similar size. The I6 markers closest to the resistance locus were cloned and secluenced. The W07:h marker. which showed no recombination with the rejistance locus but is difficult to score. includes a CT rnicrosatellite repeat. The jequznce information will allow the design of SCAR primers and eventual rn~~p-hased cloning of the resistance allele. Coinrnun~c~ied hq D B. Ycale S. X. .Clehienbacher i:<j R. N. Brown J . Resistance Gene Analog Polymorphism (RGAP) Markers Co-Localize with the Major QTL of Fusarium Head Blight (FHB) Resistance, Qfhs.ndsu-3BS in Wheat

Resistance gene analog polymorphism (RGAP) markers linked to Fusarium head blight resistance (FHB) and co-localize with Qfhs.ndsu-3BS were identified using F3 plants and F3:5 lines derived from a ‘Wangshuibai’ (resistant) / ‘Seri82’ (susceptible) cross. The mapping populations were genotyped using 50 degenerate primers designed based on the known R genes. Out of the 50 designed primer combinati...

Identifícation of RAPD markers linked to bacterial blight resistance genes in Phaseolous vulgaris L

Genetic studies indicate that resistance to bacterial blight is different depending on the source of resistance may be determined by both major and minor genes (Mcelroy 1985, Musaana el al, 1993. ). RAPD markers obtained from PCR amplification of genomic DNA with random primers have been successfully used for tagging genes of interest in common bean and other crops. For seed, pod and leaf react...

RAPD based DNA markers linked to anthracnose disease resistance in Sorghum bicolor ( L.) Moench.

Anthracnose caused by Colletotrichum graminicola is one of the major diseases of sorghum. The locus for disease resistance in sorghum [Sorghum biocolor (L.) Moench] accession G73 was found to segregate as a simple recessive trait in a cross to susceptible cultivar HC136. In order to identify molecular markers linked to the locus for disease resistance, random amplified polymorphic DNA (RAPD) an...

IDENTIFICATION OF MOLECULAR MARKERS LINKED TO LEAF CURL VIRUS DISEASE RESISTANCE IN COTTON

The identification of molecular markers linked to leaf curl virus (CLCuV) disease resistance in cotton has the potential to improve both the efficiency and the efficacy of selection in cotton breeding programs. Genetic analysis suggested that CLCuV resistance is controlled by a single dominant gene. In this study, an interspecific F2 population derived from a cross of Gossypium barbadense and G...

Virulence and Genetic Variability Among Isolates of Mycosphaerella pinodes

Mycosphaerella blight, caused by Mycosphaerella pinodes (Berk. & Bloxam) Vestergr., is the most important foliar disease of field pea (Pisum sativum L.) worldwide (2,7,16,17,23,30,31). In Canada, average dry seed yield losses caused by this disease have been estimated at 10%, and losses of over 50% have been reported in field trials (28,34). Management for Mycosphaerella blight is limited to fu...
